PolySULFOETHYL A - for Cation-Exchange of Peptides - This strong cation-exchange (SCX) material was developed specifically for HPLC of peptides. At pH 2.7-3.0, peptides lose their (-) charges and have net (+) charge. Thus, PolySULFOETHYL Ais a general-purpose alternative to reversed-phase (RPC), fractionating peptides by differences in charge rather than polarity. Compared to other SCX materials based on sulfopropyl- (SP-) groups, PolySULFOETHYL A is unusually hydrophilic. This minimizes hydrophobic interactions with peptides, with high recovery and less peak tailing. Capacity is also high, permitting better retention and fractionation of the weakly basic peptides from tryptic digests. The capacity of an ion-exchange material like this is 4x that of a comparable RPC column. Therefore, ion-exchange should be the initial step of a multi-step purification. Use PolySULFOETHYL A for: 1) Multidimensional HPLC of peptide mixtures, such as tryptic digests in proteomics analyses (including iTRAQ and ICAT reaction products). 2) Isolation of peptides from natural products. 3) QC and purification of synthetic peptides. 4) Selective isolation of disulfide-linked peptides, phosphopeptides and C-terminal fragments from tryptic digests. 5) Mapping of peptide digests (tryptic, V8, CNBr, etc.). Proteins can be run on PolySULFOETHYL A columns too; at pH 3, retention is all but guaranteed. An example is the analysis of Lung Surfactant Protein on a PolySULFOETHYL A column operated in the Hydrophilic Interaction (HILIC) mode. Standard material for peptide applications is 300-Å, in either 3- or 5-µm. The 200-Å material has about 25% greater capacity and is preferred for phosphopeptide isolation and fractionation of iTRAQ® reaction mixtures. For proteins, use 1000-Å material or the 3-µm, 1500-Å material.HIC materials separate proteins on the basis of hydrophobic character, as does RPC. However, HIC uses totally aqueous buffers, retaining tertiary structure and biological activity. Selectivity is generally superior to that of RPC for proteins and polypeptides large enough to have significant secondary or tertiary structure. Typically, a sample is eluted with a decreasing gradient of a salt such as a sulfate or phosphate. Proteins elute in order of increasing surface hydrophobicity. Surfactants (e.g., CHAPS; octylglucoside) can be added to the mobile phase if necessary. The relative hydrophobic character of PolyPROPYL A, PolyETHYL A and PolyMETHYL A is 100, 60 and 15, resp. HIC has greater sensitivity than other modes to the location of a modification or residue difference, especially (but not necessarily) if it involves a nonpolar group. Use these HIC materials for: 1) Multidimensional protein purification (suggested sequence: Ion-exchange - [add salt to collected fractions] - HIC). 2) Purification of polypeptides (e.g., venoms and glycopeptides). 3) Characterization of antibodies. 4) QC analysis of proteins differing in the polarity of a single residue or modified positional variants. Use material with 1000- or 1500-Å pores for proteins > 20 KDa. Capacity is comparable to ion-exchange. Unless a protein is known to be unusually hydrophobic, use PolyPROPYL A.PolyGLYCOPLEX A - for HILIC of Complex Carbohydrates - This neutral material has the unusually high capacity needed to retain complex carbohydrates in the HILIC mode. Columns can frequently be operated with just acetonitrile and water, although charged carbohydrates may require inclusion of some salt such as ammonium acetate. These conditions are convenient for isolation of carbohydrates or direct flow to a mass spectrometer. Selectivity is good for both native glycans and derivatives such as those with the 2-aminopyridine (PA-) fluorophore. Oligosaccharide mixtures can often be resolved isocratically, although gradients are recommended for especially varied samples. Sialylated and asialo- glycans can be resolved using the same running conditions. In some cases selectivity compares favorably with that of HPAEC with PAD detection, and both running conditions and equipment maintenance are far more convenient.

Such unusual pH-resistance values have been secured as a result of phase bonding efficiency and a proprietary endcapping process which provides a protective shield that impedes acidic and basic eluents from attacking the silica surface. Until quite recently, silica packings were limited to working between pH 2 and pH 7 since below pH 2 the bonds between the C18 chains and the silica particle were hydrolysed, resulting in a gradual loss of retention capacity of the column. Above pH 7 the problem that arose was one of simply dissolving the silica, and therefore the pure destruction of the column. These are no longer a problem thanks to Cronusil-M. An eluent of ACN/TFA 1% pH 1.0 (10:90) 1ml/min 25°C is passed through the column at regular periods, checked with the reverse phase test and a retention comparison is made of the last anthracene peak.An eluent of ACN//1-methylpyrrolidine 50mM pH 11.5 50:50, 1ml/min 25°C is passed through the column. With the same eluent 10ml of diphenhydramine (1mg/ml dissolved in water) is injected and the efficiency and symmetry of the peak is tested. The pH stability graphs above show the efficiency of the process. Eluting the Cronusil-M HPLC column for 78 hours at pH 11.5, showed no significant deterioration in terms of both efficiency and peak symmetry for diphenhydramine. With an eluent as acidic as pH 1, the column stabilises in a short period of time making it possible to work even in these extreme conditions.
